5′-tert-Butoxycarbonylamino-6′-(3-chloro-benzyloxy)-2,3,5,6-tetrahydro-[1,2′]bipyrazinyl-4-carboxylic acid tert-butyl ester I-3q (50 mg, 96 μmol) was dissolved in CH2Cl2 (1 mL) and trifluoroacetic acid (1 mL) was added with stirring at room temperature. The solution was stirred at room temperature for 3 h then diluted with CH2Cl2 (20 mL). The CH2Cl2 solution was washed with saturated aqueous NaHCO3 (3×50 mL) and brine (25 mL), dried over Na2SO4 (anhydrous), filtered, and concentrated in vacuo to give the title compound 3-O as a reddish-brown solid (29 mg, 95% yield).
