7-Amino-2,4-dioxo-1-[4-(4-diphenylmethoxypiperidino)butyl]-1,2,3,4-tetrahydroquinazoline (700 mg) obtained in Example 39 was dissolved in tetrahydrofuran (10.0 ml), and triethylamine (0.24 ml) and ethyl malonyl chloride (0.20 ml) were added with stirring and cooling on ice. The reaction mixture was stirred at room temperature for 18. 5 hours and combined with water and extracted with ethyl acetate. After the extract was washed with water and dried (Na2SO4), the solvent was distilled off under reduced pressure to give the title compound (690 mg, 80%) as an amorphous.
