Reaction Steps

Ethyl 3-[4-(5-{5-chloro-6-[(1-methylethyl)oxy]-3-pyridinyl}-1,2,4-oxadiazol-3-yl)-1H-indazol-1-yl]-2,2-dimethylpropanoate (D105) (50 mg, 0.103 mmol) was dissolved in ethanol (10 ml). Sodium hydroxide (2N in H2O, 0.258 ml, 0.517 mmol) was added and the resulting mixture was stirred at 60° C. overnight then cooled to room temperature. Solvent was evaporated, then the residue was diluted with water. The aqueous phase was neutralised with a 2N HCl aqueous solution then extracted with EtOAc. The organic phase was washed with H2O, dried over MgSO4 and concentrated in vacuo to give the title compound (22 mg, 42%) as an off-white solid. MS (ES): C22H22ClN5O4 requires 455; found (MH+) 456.
