Reaction Steps

To a mixture of tert-butyl 4-(3-amino-1,2,4-thiadiazol-5-yl)phenylcarbamate (0.078 g, 0.27 mmol), DMAP (0.049 g, 0.40 mmol), and CDI (0.066 g, 0.41 mmol) was added DMF (1 mL). The reaction mixture was heated to 50° C. for 15 h and 3,3-diphenyl-N-(2-(pyridin-2-yl)ethyl)propan-1-amine (0.090 g, 0.28 mmol) was added. The reaction mixture was heated to 50° C. for 20 h. Direct purification by flash column chromatography on silica gel (eluted with 50% to 100% EtOAc in hexanes) gave tert-butyl 4-(3-(3-(3,3-diphenylpropyl)-3-(2-(pyridin-2-yl)ethyl)ureido)-1,2,4-thiadiazol-5-yl)phenylcarbamate 156.
