Reaction Steps
Trimethylaluminum (2.0 M in toluene, 1.2 mL, 2.4 mmol) was added to a toluene (8 mL) solution of aniline (248 mg, 2.66 mmol). After stirring at room temperature for 2 hours, solid ethyl 4-amino-2-[6-chloro-1-(3,3,4,4,4-pentafluorobutyl)-1H-indazol-3-yl]-5-methyl-6-oxo-6,7-dihydro-5H-pyrrolo[2,3-d]pyrimidine-5-carboxylate, as described in Example 157, (250 mg, 0.469 mmol) was added. The reaction solution was stirred at 50° C. for 30 minutes and then at room temperature overnight. Sodium potassium tartrate (0.5M aqueous, 25 mL) and EtOAc (25 mL) were added and the mixture stirred vigorously for 30 minutes. The phases were separated and the organic phase was washed with water (2×) and brine. The organic phase was then dried over MgSO4, filtered and concentrated in vacuo. The residue was purified by silica gel chromatography using a hexanes/EtOAc gradient to give the indicated product. Chiral separation using SFC on a Chiralpak AD-H column provided both enantiomers of the title compound. 1H NMR (400 MHz, CH3CN-d3): δ 9.19 (s, 1H); 8.55 (d, J=8.69 Hz, 1H); 7.64 (s, 1H); 7.55 (d, J=8.01 Hz, 2H); 7.32 (t, J=7.77 Hz, 2H); 7.20-7.10 (m, 2H); 6.45 (s, 2H); 4.72 (t, J=7.10 Hz, 2H); 2.85-2.69 (m, 2H); 1.79 (s, 3H). m/z=580.2 (M+H).
