Reaction Steps

To an ice-cooled suspension of 6-bromo-1H-pyrrolo[3,2-b]pyridine (7.0 g, 35 mmol) in N,N-dimethylformamide (100 mL) was slowly added a solution of sodium hydride (2.12 g, 88.28 mmol, 60% in mineral oil) in N,N-dimethylformamide (20 mL). After 30 min at 0° C., a solution of 2-(1-chloroethyl)pyridine (5.0 g, 35 mmol) in N,N-dimethylformamide (20 mL) was slowly added. The mixture was then warmed to room temperature. After 16 h, saturated aqueous ammonium chloride solution (20 mL) was slowly added to the reaction mixture, and the resulting solution was concentrated in vacuo. The resulting residue was purified by flash column chromatography (10→30% ethyl acetate in petroleum ether) to afford 6-bromo-1-(1-(pyridin-2-yl)ethyl)-1H-pyrrolo[3,2-b]pyridine (6.0 g, 56% yield). 1H NMR (400 MHz, Chloroform-d) δ 8.61 (d, J=4.4 Hz, 1 H), 8.47 (d, J=2.0 Hz, 1 H), 7.70 (s, 1 H), 7.62-7.57 (m, 2 H), 7.21-7.18 (m, 1 H), 6.81-6.76 (m, 2 H), 5.66-5.61 (m, 1 H), 2.00 (d, J=7.2 Hz, 3 H).
