Reaction Steps

To a solution of 4-(1-(2,6-difluorophenyl)-4-methoxy-1H-pyrazolo[4,3-c]pyridin-3-yl)-N-methylbenzamide (2.68 g) in acetonitrile (50 mL) were added sodium iodide (2.04 g) and chloro(trimethyl)silane (4.34 mL) was added, and the mixture was stirred at 60° C. for 3 hr. To the reaction mixture was added saturated aqueous sodium hydrogencarbonate solution, and the mixture was extracted with ethyl acetate. The organic layer was washed with saturated brine, dried over anhydrous sodium sulfate, and concentrated under reduced pressure. The residue was purified by silica gel column chromatography (basic silica gel, hexane/ethyl acetate to ethyl acetate/methanol). 1.60 g of the obtained title compound (2.15 g) was dissolved in 2-propanol (240 mL) at 80° C. To the solution was added heptane (160 mL) over 15 min, the mixture was stirred for 15 min, and allowed overnight to be cooled. The white precipitate was collected by filtration, washed with a mixed solvent of 2-propanol-heptane (3:4), and dried in vacuum to give the title compound (1.49 g).
