Reaction Steps
Methyl 2-chloro-6-(4-{[(3,4-dichloro-5-methyl-1H-pyrrol-2-yl)carbonyl]amino}piperidin-1-yl)pyrimidine-4-carboxylate (Example 6) (500 mg, 1.12 mmol), hydrazine (0.035 ml, 1.12 mmol), and TEA (0.16 ml, 1.12 mmol) were combined in DMF (3 ml) and stirred at room temperature. Within 1.5 h, a white precipitate had formed. The reaction was stirred for another 30 minutes, and then the precipitate was collected by suction filtration to give 377 mg of the title compound. 100 mg of the crude material was purified by preparatory HPLC, using a gradient of 40-70% acetonitrile/water (0.1% TFA) over 14 minutes to give 23 mg of the title compound.
