Reaction Steps
2.68 g 2-(6-Chloro-3-methoxy-pyridazin-4-yl)-6-formyl-indole-1-carboxylic acid tert-butyl ester is dissolved in 300 mL dichloromethane. 4.73 mL N-methylpiperazine and then 30 mL acetic acid are added. 4.40 g sodium triacetoxyborohydride is added in several portions over 3 hours. Sat. aq. NaHCO3-solution is added and the reaction mixture is stirred until the CO2-evolution stopped. The reaction mixture is diluted with dichloromethane, the aq. phase washed with dichloromethane, the combined org. phases dried (MgSO4) and concentrated in vacuo. HPLC purification affords 2.019 g (50%) 2-(6-chloro-3-methoxy-pyridazin-4-yl)-6-(4-methyl-piperazin-1-ylmethyl)-indole-1-carboxylic acid tert-butyl ester as its trifluoro-acetate salt.
