Reaction Steps
1,1-Dimethylethyl (2R,6S)-4-[6-{[(4-bromo-2-chlorophenyl)sulfonyl]amino}-5-(methyloxy)-2-pyridinyl]-2,6-dimethyl-1-piperazinecarboxylate (D13) (0.10 g, 0.169 mmol), 5-methylthiophen-2-boronic acid (0.0739 g, 0.254 mmol), Palladium dichloride di-triphenylphosphine (5.9 mg, 0.0127 mmol), sodium carbonate (0.0359 g, 0.339 mmol) were heated in DME (1 mL) and water (0.5 mL) at 120° C. in the microwave for 20 minutes. The reaction was then diluted with ethyl acetate (50 mL) and washed with saturated sodium hydrogen carbonate (2×30 mL) and brine (30 mL). The organic layer was dried (MgSO4) and evaporated to give a yellow foam which was purified by chromatography [silica gel, eluting with 0 to 70% Ethyl acetate/pentane]. Product fractions evaporated to give a pale yellow oil (D16) (0.088 g) MS (ES+) m/e 607/609 [M+H]+.
