Reaction Steps
A solution of 3-bromo-benzothiophene (commercially available, 0.232 g) and 4-(4-(2-methoxyphenyl)piperazin-1-yl)piperidine (Step 2, 0.30 g) in tetrahydrofuran was added sodium tert-butoxide (0.15 g), tris-(dibenzylideneacetone)dipalladium(0) (Pd2(dba)3, 0.10 g) and 2-dicyclohexylphosphino-2′-(N,N-dimethylamino)-biphenyl (CYMAP, 0.01 g). The reaction was heated at reflux for 24 hours. It was then cooled and filtered through celite. The filtrate was evaporated and purified by flash chromatography on silica gel using ethyl acetate/hexanes to give 0.10 g of the desired product. MP: 156-158° C.; MS (ES) m/z (relative intensity): 408 (M+H)+ (100).
