Reaction Steps
1-(3-Dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (EDCL HCl) (1.0 mmol) is added to a mixture of 4-(3-{[4-(3-methyl-5-oxo-4,5-dihydro-1H-1,2,4-triazol-1-yl)phenyl]thio}phenyl)tetrahydro-2H-pyran-4-carboxylic acid (1.0 mmol) (example 15), 1-hydroxybenzotriazole (1.0 mmol), N-methylmorpholine (1.65 mmol) and propargylamine (1.0 mmol) in dichloromethane-dimethylformamide (1:1) at 0° C. The solution is stirred overnight at room temperature, washed with water and the aqueous layer is extracted with dichloromethane. The combined organic extracts are dried with sodium sulphate, filtered and the solvent is evaporated under reduced pressure. The residue is purified by column chromatography over silica gel to yield the title product.
