Reaction Steps
The compound 1,1-dimethylethyl 4-{[8-(2,6-difluorophenyl)-4-(2-methyl-5-{[(phenylmethyl)amino]carbonyl}phenyl)-7-oxo-5,6,7,8-tetrahydropyrimido[4,5-d]pyrimidin-2-yl]amino}-1-piperidinecarboxylate, (0.052 g, 0.0076 mmol) was dissolved in CH2Cl2 (4 mL) and stirred under argon at 0° C. Trifluoroacetic acid (1 mL) was added and the mixture stirred under argon at 0° C. for 2 h. The solvents were pumped off in vacuo, and the residue was partitioned between EtOAc and H2O. The mixture was made basic by the addition of 1 M NaOH. The organic phase was washed with H2O (2×), brine (1×), dried over anhydrous Na2SO4, filtered and evaporated. The residue was flash chromatographed on silica gel (10 g) eluted with 6:2:0.2, CH2Cl2:ethanol:NH4OH to give the title compound as a white amorphous solid. mp (dec) 172-176° C.; LC-MS m/z 584 (M+H)+, 1.49 min (ret time).
