Reaction Steps

4-Methylmorpholine (0.90 mL, 8.19 mmol) was added to a suspension of 2-(1H-benzotriazole-1-yl)-1,1,3,3-tetramethyluronium hexafluorophosphate (HBTU, 1.55 g, 4.10 mmol), 1-hydroxybenzotriazole monohydrate (627 mg, 4.10 mmol) and 4-(4-propylpiperazin-1-yl)benzoic acid (85% wt., 1.25 g, 4.29 mmol) in dimethylformamide (6 mL). The suspension was shaken for 5 minutes then added to a stirred solution of (S)-2-amino-1-((3R,3aR,6aR)-3-hydroxydihydro-2H-furo[3,2-b]pyrrol-4(5H,6H,6aH)-yl)-4-methylpentan-1-one hydrochloride (prepared as above, assume 3.90 mmol) in dimethylformamide (10 mL). Residual material was washed into the reaction flask using dimethylformamide (2×2 mL). After 2.5 hours the majority of solvents were removed in vacuo. The residue was dissolved in dichloromethane (100 mL) and washed with saturated sodium hydrogen carbonate solution (75 mL). The aqueous phase was extracted with dichloromethane (2×50 mL) then the combined organic layers washed with brine (50 mL), then dried (Na2SO4), filtered and reduced in vacuo. Flash chromatography over silica, eluting with heptane:ethyl acetate mixtures 100:0:0 to 100 then methanol:dichloromethane mixtures 1:99 to 5:95 gave N—((S)-1-((3R,3aR,6aR)-3-hydroxydihydro-2H-furo[3,2-b]pyrrol-4(5H,6H, 6aH)-yl)-4-methyl-1-oxopentan-2-yl)-4-(4-propylpiperazin-1-yl)benzamide as an off-white solid (1.67 g, 91%).
