Reaction Steps
Under a nitrogen gas stream, to a solution of 4-(3-phenyl-1,2,4-thiadiazol-5-yl)-N-pyridin-3-ylpiperazine-1-carboxamide (100 mg, 0.273 mmol) in dimethylformamide (1.0 ml) was added, under ice-cooling, sodium hydride (13.1 mg, 0.327 mmol), and the mixture was stirred at room temperature for 30 minutes. Methyl iodide (0.020 ml, 0.327 mmol) was added thereto under ice-cooling, and the mixture was stirred at 0° C. for 30 minutes, followed by stirring at room temperature for 2 hours and half. An aqueous saturated ammonium chloride solution was poured to the reaction mixture, and the resulting solution was extracted with ethyl acetate. The extract was washed with water and dried over anhydrous magnesium sulfate, and the solvent was distilled off under reduced pressure. The residue was purified by silica gel column chromatography (ethyl acetate) and recrystallized from a mixed solvent of hexane and ethyl acetate to give 43.0 mg (41.4%) of the desired product as a solid.
