Reaction Steps
4-Methyl-6-chloropyrimidine (0.51 g) was added to a stirred mixture of 1-(4-bromophenylsulphonyl)-4-(1-piperazinylcarbonyl)piperazine hydrochloride (1.50 g) and ethanol (35 ml). Triethylamine (2.2 ml) was added and the mixture was heated at reflux for 6 hours. The solvent was removed by evaporation and the residue was purified by chromatography on silica gel using a gradient of 0% to 10% methanol in dichloromethane as eluent to give a foam. This was crystallised from methyl tert-butyl ether to give 1-(4-bromophenylsulphonyl)-4-[1-(4-methylpyrimidin-6-yl)piperazin-4-ylcarbonyl]piperazine as a solid (0.77 g); m.p. 215-216° C.; NMR (CDCl3): 8.5 (s, 1H), 7.7 (m, 2H), 7.6 (m, 2H), 6.35 (s, 1H), 3.6 (m, 4H), 3.4 (m, 4H), 3.3 (m, 4H), 3.05 (m, 4H), 2.33 (s, 3H).
