Reaction Steps
A mixture of 2-trifluoroacetylamino-N,N,3-trimethylbenzamide (584.2 mg, 2.13 mmol), prepared as in Example 22, and sodium hydride (110 mg, 2.7 mmol) in 10 mL of DMF was heated at 50° C. for 20 minutes and then 1-chloro-3-[4-(2-methoxyphenyl)piperazin-1-yl]propane (647 mg, 2.13 mmol) was added. The mixture was heated at 80° C. for 18 hours, then poured into water and extracted with ethyl acetate (3×50 mL). The combined extract was washed with saline, dried (MgSO4) and concentrated. The residue was purified on silica gel by column chromatography eluting with 6% methanol/methylene chloride to give 2-{3-[4-(2-methoxy-phenyl)piperazin-1-yl]propylamino}-N,N,3-trimethylbenzamide (100 mg, 0.24 mmol).
