Reaction Steps
Potassium cyanate (4.9 g) is suspended in dichloromethane (50 ml) followed by dropwise addition of trifluoroacetic acid (4.4 ml) at 0° C. A solution of 3-[1-(2-aminoethyl)piperidin-4-yl]-5-chloro-1-(4-fluorophenyl)- 1H-indole (10.8 g) prepared according to Example 1 in dichloromethane (100 ml) is added dropwise followed by stirring for 6 hours at room temperature. Water (100 ml) is added and the reaction mixture is made alkaline with concentrated ammonia. The phases are separated, followed by extraction with dichloromethane. The combined organic phases are dried over magnesium sulfate. Removal of solvent under vacuum gives a heavy oil which is purified by flash chromatography (silica gel, eluent: triethylamine/methanol/ethyl acetate 5:20:75). About 7.9 g of the title compound is isolated as a crystalline material.
