Reaction Steps
6.0 g of 5-chloro-4-{2-[4-(2,3-dihydroxypropyl)-2-methylphenoxy]ethylamino}-6-methylpyrimidine and 50 ml of acetone were dissolved in 200 ml of toluene and a small amount of p-toluenesulfonic acid was added to the solution. The mixture was then heated under reflux for 6 hours. At the end of this time, the reaction mixture was adjusted to weakly alkaline by the addition of a dilute aqueous solution of sodium hydroxide. Then the reaction mixture was washed with water and dried over anhydrous sodium sulfate. Toluene was removed by distillation under reduced pressure. The oily product thus obtained was subjected to column chromatography (Wakogel C-200, eluted with a 2:1 by volume mixture of toluene and ethyl acetate) to isolate the product and give 4.8 g of the title compound as a pale yellow oily liquid, n25.0 =1.5516.
