Procedure
A solution of 5-acetylmercaptomethyl-2-methyltetrazole (0.400 g, 2.32 mmol) in dry dichloromethane (3 mL) was treated with methyltriflate (0.76 g, 4.64 mmol) and stirred at 22° C. for 16 h. Evaporation of the solvent under vacuum gave a red oil. This salt was dissolved in cold oxygen-free water (5 mL) and treated with 4M sodium hydroxide (0.8 mL, 3.2 mmol). The mixture was stirred at 0° C. for 40 min, diluted with water (7 mL), and the pH was adjusted to 7.3 with saturated KH2PO4. The clear resulting solution was maintained under nitrogen and used immediately for the following step.
