7-Isobutyl-5-methyl-3-(phenylamino)-2-(4-(pyridin-2-yl)benzyl)-4-thioxo-4,5-dihydro-2H-pyrazolo[3,4-d]pyrimidin-6(7H)-one (15 mg, 0.030 mmol) and HgCl2 (16.4 mg, 0.060 mmol) are suspended in THF, and then 2.0 M methylamine solution in THF (240 μL, 0.12 mmol) is added. The reaction mixture is heated in a Biotage microwave at 110° C. for 5 hours. After routine workup, the mixture is purified by a semi-preparative HPLC to give pure product as white solids. MS (ESI) m/z 494.3 [M+H]+.
