Procedure
Lithium aluminum hydride was added in portions to a stirring solution of 2-amino-5-methyl-7-[3-(trifluoromethyl)phenyl]imidazo[5,1-f][1,2,4]triazin-4(3H)-one (Intermediate 43) (2.0 g, 6.47 mmol) in DME (50 mL) at rt. The resultant mixture was then heated to reflux for 2 h. The reaction mixture was then cooled to 0° C. and carefully quenched with H2O. After extraction into EtOAc., the combined organic extracts were dried (MgSO4), filtered, and concentrated. Purification of the residue by chromatography afforded 2-methyl-7-[3-(trifluoromethyl)phenyl]-3,4-dihydroimidazo[5,1-f][1,2,4]triazin-2-amine (0.60 g) as a pale yellow solid. 1H NMR (DMSO-d6): δ8.67-8.60 (m, 1H), 8.54 (s, 1H), 7.61-7.54 (m, 2H), 6.55 (s, 1H), 5.50 (s, 2H), 4.35 (s, 2H), 2.06 (s, 3H). MS m/z 294 (M−1).
