A solution of N-{2-[4-methoxy-3-({5-methyl-7-[3-(trifluoromethyl)phenyl]-imidazo[5,1-f][1,2,4]triazin-2-yl}amino)phenyl]ethyl}acetamide (Example 52) (0.050 g, 0.17 mmol) in dilute HCl(aq) (1N, 15 mL) was heated to reflux for 5 h. Neutralization with NaOH(aq) (1N) and extraction into EtOAc followed by concentration and preparative HPLC gave N-[5-(2-aminoethyl)-2-methoxyphenyl]-5-methyl-7-[3-(trifluoromethyl)-phenyl]imidazo[5,1-f][1,2,4]triazin-2-amine (0.013 g) as a yellow solid. 1H NMR (Acetone-d6): δ9.17 (s, 1H), 9.08 (d, J=8.1 Hz, 1H), 8.64 (s, 1H), 8.33-8.30 (m, 1H), 8.13 (s, 1H), 7.97-7.90 (m, 1H), 7.80-7.77 (m, 1H), 7.02-6.95 (m, 2H), 3.96 (s, 3H), 3.48 (t, J=7.4 Hz, 2H), 2.95 (t, J=7.4 Hz, 2H), 2.60 (s, 3H). MS m/z 443 (M+1).
