Reaction Steps
To 4,4′-dimethoxybenzhydrol (5.62 g, 23.0 mmol) was carefully added thionyl chloride (20.1 mL, 276 mmol) and the mixture was heated to reflux for about 1 hour. The solution was cooled, evaporated to dryness and the residue was dissolved in tetrahydrofuran (50 mL). This solution was then added to a solution of Example 326 (5.3 g, 14.0 mmol) and triethylamine (5.9 mL, 42.0 mmol) in tetrahydrofuran (100 mL). The reaction mixture was heated to about 50° C. for about 2.5 hours, cooled, diluted with water and extracted with ethyl acetate. The combined organic extracts were dried (Na2SO4), filtered and concentrated under vacuum, and the residue was purified by flash chromatography on silica gel using dichloromethane/methanol (10:1) as eluent to provide the desired product. MS (ESI): m/z 605 (M+H)+.
