Reaction Steps
A solution of commercially available Z-Pro-OH (3.9 g, 11.0 mmol) was dissolved in dry dichloromethane (25 ml) and cooled in an ice bath. HBTU (4.2 g, 11.0 mmol) was added followed by HOBt.H2O (1.68 g, 11.0 mmol) and DIEA (3.12 g, 24.2 mmol). The reaction mixture was stirred for 10 min and then to the above activated acid, H-Arg(Pmc)-t-Bu (5.0 g, 10.0 mmol) was added as a solid. The solution was allowed to come to RT and stirred for 20 h at RT. The mixture was poured into saturated sodium bicarbonate solution (100 mL) and stirred for 5 min. The organic layer was separated and washed with water (2×100 mL) and dried (sodium sulfate). Evaporation of the solvent yielded the product as colorless foam. Yield: 8.2 g (100%).
