Reaction Steps
A solution of n-BuLi (2.5 M in hexanes, 5 mL, 12 mmol) was added dropwise into 2-(1,4-dioxa-spiro[4.5]dec-8-yl)-thiazole (2.50 g, 11 mmol) in THF (10 mL) at −78° C. The reaction was kept at −78° C. for 30 min. The solution was added to solid dry ice (˜5 g) dropwise. After addition, the reaction was warmed to room temperature slowly over 1 hour and quenched with 1 N NaOH (˜10 mL) solution. The solvent was removed and the residue was partitioned between ether and water. The aqueous layer was adjusted to pH=6 with 5% HCl and extracted with ethyl acetate three times. The combined organic layers were washed with brine, dried over anhydrous Na2SO4, filtered and concentrated to give the title compound as white solid.
