A mixture of 6-tert-butylfuro[2,3-d]pyrimidin-4(3H)-one (1.0 g) and POCl3 (10 mL) was stirred at 55-65° C. for 3 h. The reaction mixture was poured into ice cold water and stirred for 10 min. The solution was extracted with chloroform (3×100 mL) and the combined chloroform layer was washed with aq. NaHCO3 solution, water, brine and dried over sodium sulfate. The solution was filtered and evaporated the solvent. The residue was chromatographed over silica gel column using hexane-EtOAc (90:10) as eluents to give the product as a white color solid (800 mg, 73%), mp 72-74° C. 1H NMR (400 MHz, CDCl3): δ 8.69 (1H, s), 6.48 (1H, s), 1.42 (9H, s).
