Reaction mixture in 100 mM phosphate buffer, pH 7.4, (total volume, 200 μl) was incubated at 37° C. for 30 minutes. The reaction mixture contained a combination of cytochrome c (40 μg), ovine prostaglandin H2 synthase form 2 (104 units), arachidonic acid (590 μM), epinephrine (31 μg), hematin (1 μM), and DETA NONOate, an NO donor, (300 μM) as indicated in FIG. 2. After incubation, the proteins were separated by SDS-PAGE followed by Western blot analysis with an alkaline phosphatase system using monoclonal antibodies produced against nitrotyrosine.
