A solution of 0.764 g (1.41 mmol) of methyl-4-{[4-({6-[(tert-butoxycarbonyl)amino]-2-hexynyl}oxy)pheny]sulfonyl}-2,2-dimethyl-3-thiomorpholinecarboxylate and 3.77 g (28.2 mmol) of lithium iodide in 30 mL of ethyl acetate was heated to reflux for 18 h. The resulting reaction was acidified, washed with water, an aqueous solution of Na2S2O3, brine, dried over MgSO4, filtered and concentrated in vacuo. The residue was chromatographed on silica gel eluting with 0.5% methanol/dichloromethane to provide 0.417 g (56%) of 4-{[4-({6-[(tert-butoxycarbonyl)amino]-2-hexynyl}oxy)phenyl]sulfonyl}-2,2-dimethyl-3-thiomorpholine carboxylic acid as a white solid.
