Reaction Steps
Sodium hydride (60% in mineral oil, 16.6 mg, 0.312 mmol) was added to a solution of tert-butyl 1,3,4,8-tetrahydro-2H-[1,4]oxazepino[6,7-e]indole-2-carboxylate (Intermediate 18, 0.060 g, 0.21 mmol) in DMF (3 mL). The mixture was stirred for 20 minutes at room temperature before (2-methoxy-5-methylphenyl)sulfonyl chloride (69 mg, 0.31 mmol) was added. The mixture was stirred for 20 minutes and diluted with water and DCM. The aqueous phase was adjusted to pH 3 with 1 M HCl and extracted with DCM (2×). The combined organics were evaporated to approximately 5 mL. TFA (2 mL) was added and the mixture was stirred at room temperature for 3 hours and the reaction mixture was evaporated. The residue was dissolved in DCM and triethylamine (1 mL) was added and the mixture was evaporated. The crude product was purified by flash chromatography on silica gel using MeOH:DCM:NEt3 (3:96:1) as the eluent to give a white solid which was suspended in diethylether and filtered. The white powder was washed with cold diethylether (2×) and dried in vacuo to give the title compound (65 mg). MS m/z 373 [M+H]+.
