Reaction Steps
To a solution of 2-((S)-1-tertbutoxycarbonylaminoethyl)-5-fluoro-3-phenyl-3H-benzoimidazole-4-carboxylic acid methyl ester (1.1 g, 2.7 mmol) in MeOH (20 mL) was added 4N HCl in dioxane (5 mL) and the reaction mixture was heated at 45° C. for 3 h. The volatiles were removed under reduced pressure and the resulting residue was treated with 6-chloro-9-(tetrahydropyran-2-yl)-9H-purine (825 mg, 3.46 mmol) and DIPEA (1.8 mL, 10.6 mmol) in n-butanol (10 mL). The reaction mixture was heated in a sealed vial for 16 h at 90° C. After cooling to RT, the volatiles were removed under reduced pressure and the resulting residue purified by column chromatography (Si—PCC, gradient 0-10% MeOH in EtOAc) to afford the title compound (1.2 g, 87%). LCMS (Method C): RT 3.11 min [M+H]+ 516.2.
