Procedure
Potassium hydroxide (1.42 g, 25.4 mmol) was added to a solution of methyl 6-(methylsulfonyl)-3-{[4-(4-morpholinyl)-1-piperidinyl]methyl}-2-[3-(trifluoromethyl)phenyl]-4-quinolinecarboxylate (3.0 g, 5.07 mmol) in methanol (60 mL) and water (20 mL), and the resulting mixture was heated to reflux for 5 h. The solvent was removed under reduced pressure, and the residue was adjusted to pH˜5-6 with 2N HCl and extracted with methylene chloride (three times). The combined organic extracts were washed with brine, dried over Na2SO4, filtered, and concentrated in vacuo to give 6-(methylsulfonyl)-3-{[4-(4-morpholinyl)-1-piperidinyl]methyl}-2-[3-(trifluoromethyl)phenyl]-4-quinolinecarboxylic acid (2.73 g, 81% yield). MS (m/z) 578.2 (M+H+).
