Reaction Steps
N-[(t-Butyloxycarbonyl)-pyrrolidin-2(S)-ylmethyl]-3(S)-ethyl-proline (0.50 g, 1.53 mmol), EDC (0.293 g, 1.53 mmol), HOBT (0.243 g, 1.53 mmol) and 3-chlorobenzylamine(0.187 mL, 1.53 mmol) were dissolved in DMF (5 mL), the pH adjusted to 7 with N-methyl-morpholine (0.51 mL, 4.6 mmol), and the reaction mixture stirred for 18 hours at ambient temperature. After removing the solvent in vacuo the residue was partitioned between EtOAc and 5% aqueous NaHCO3. The organic layer was washed with brine, dried (MgSO4), filtered, and concentrated to give the title compound after chromatography (SiO2, EtOAc:hexane, 2:3).
