Reaction Steps

Step 1): To a solution of 2amino-5nitro-N-[(1-diphenylmethylpiperidin-4-yl)methyl]benzamide (Preparation 28) (1.2 g, 2.69 mmol) in CHCl3 (15 ml) was added n-butylisocyanate (802 mg, 8.09 mmol). After the solution was refluxed for 24 hours, the solvent was evaporated. The residue was purified by column chromatography on silica gel to give 5-nitro-2-(N'-n-butylureido)-N-[(1-diphenylmethylpiperidin-4-yl)methyl]benzamide, 1.1 g (75%).
