Reaction Steps
The crude carbamate was dissolved into MeOH (4 ml). 2 M aqueous solution of NaOH was added at room temperature. And the mixture was stirred at room temperature for 4 hours. After the addition of NH4Cl for neutrallization, the resulting reaction mixture was concentrated in vacuo. The residue was suspended into the mixed solvent of CHCl3/MeOH. The insoluble material was filtered off and the filtrate was concentrated again. The residue was purified with silica gel PTLC (eluent: CHCl3/MeOH=6/1) to give trans-5-[(tert-butoxycarbonyl)amino]-1-[4-methyl-6-(1H-pyrazol-5-ylamino)pyrimidin-2-yl]piperidine-3-carboxylic acid (10.9 mg) as colorless solid.
