Reaction Steps

To a solution of 50 mg of tert-butyl 4-[5-(2-chlorophenyl)-1-(2,6-dichlorophenyl)-2-oxo-1,2-dihydro-1,6-naphthyridin-7-yl]piperidine-1-carboxylate in 5 mL of CH2Cl2 was added 2 mL of trifluoroacetic acid. The mixture was stirred at rt for 1 h, then concentrated. The residue was dissolved in 10 mL of CH2Cl2, washed with 5 mL NaHCO3, dried (Na2SO4), and concentrated. The residue was purified by preparative thin-layer chromatography, eluting with 95:5 CH2Cl2-2M NH3 in MeOH, followed by preparative HPLC (YMC C18 20×50 mm column; 10:90 to 100:0 v/v acetonitrile/water+0.1% TFA over 5 min; 20 mL/min) to yield the title compound.
