Reaction Steps
To a stirred solution of methyl[6-chloro-2-(4-hydroxybenzoyl)-1H-indol-3-yl]acetate (Example 183, 0.15 g, 0.44 mmol) in DMF (5 ml) was added sodium hydride (15 mg, 0.46 mmol) at room temperature under nitrogen atmosphere. After 5min., 2-iodopropane (78 mg) was added and the mixture was stirred for 7 h. The mixture was quenched with 2N aqueous HCl (20 ml), and extracted with ethyl acetate (50 ml). The extract was washed with water (30 ml) and brine (30 ml), dried (MgSO4), and concentrated. The residue was purified by flash column chromatography eluting with ethyl acetate-hexane (1:2) to afford 72 mg (43%) of the title compound as white solids.
