Reaction Steps
A stirred suspension of dimethyl 3-(1-{[3-methoxy-4-(3-o-tolyl-ureido)-phenyl]-acetyl}-2,3-dihydro-1H-indol-5-yl)-pentanedioate [270 mg, Reference Example 4(c)] in ethanol (10 ml) was treated with a solution of sodium hydroxide (190 mg) in water (2 ml) and the mixture stirred at reflux for 3 hours. After cooling to room temperature the mixture was treated with water (10 ml) and then carefully acidified to pH1 by the addition of hydrochloric acid (3M) with ice cooling. the resultant precipitate was collected by filtration, then dried and then recrystallised from ethyl acetate to give the title compound (60 mg) as a white crystalline solid. LC-MS: RT=2.44 minutes (100% total area by ELS); MS (ES+) 568 (MNa+).
