Reaction Steps

3-{3-Fluoro-4-[5-(4-methyl-piperidine-1-carbonyl)-[1,2,4]oxadiazol-3-yl]-phenyl}-2-[(4′-methyl-biphenyl-3-carbonyl)-amino]-propionic acid tert-butyl ester. A mixture of 3-(4-{2-tert-butoxycarbonyl-2-[(4′-methyl-biphenyl-3-carbonyl)-amino]-ethyl}-2-fluoro-phenyl)-[1,2,4]oxadiazole-5-carboxylic acid methyl ester (50 mg) and 4-methyl-piperidine (13.5 mg) in MeOH (3 mL) was heated at 55° C. for 25 min in a microwave reactor. TLC showed the reaction was completed. The mixture was concentrated, 10 mL of 1N aq. HCl solution and 20 mL of ethyl acetate were added. The layers were separated and the aqueous portion was extracted with ethyl acetate (2×10 mL). The combined organic layers were washed with brine (2×30 mL), dried over anhydrous sodium sulfate, filtered, and concentrated to give the title compound (40 mg, 71.4% yield).
