Procedure

To a cold (−78° C.) solution of 2-((2Z)-3-(dimethylamino)-3-{2-[(4methoxy-benzyl)oxy]phenyl}-2-propenylidene)malononitrile ( 1.26 g, 3.51 mmol) in MeOH was added liquid NH3, and the mixture was heated at 120° C. in a sealed tube overnight. After cooled to room temperature, the reaction mixture was poured into water and the resulting mixture was extracted with ethyl acetate. The separated organic phase was washed with brine, and dried over MgSO4, filtered, and concentrated under reduced pressure. The crude material was purified by column chromatography on silica gel (hexane:ethyl acetate, 70:30) to give 2-amino-6-({2-[(4-methoxybenzyl)oxy]phenyl}nicotinonitrile (0.050 g, yield; 4.3%).
