Sodium hydroxide (2 N, 6 eq.) was added to a solution of methyl(7S)-7-{2-[{3-[(benzylamino)sulfonyl]propyl}(methyl)amino]ethoxy}-14-cyclohexyl-7,8-dihydro-6H-indolo[1,2-e][1,5]benzoxazocine-11-carboxylate (0.025 M) in MeOH. The reaction was heated at 60° C. for 1 h prior to introducing further sodium hydroxide (2 N, 10 eq.) and continuing heating for 2 h. The mixture was then partitioned between H2O and EtOAc. The combined organics were washed with brine, dried (Na2SO4), filtered and concentrated in vacuo. The product was used in the next step without further purification. (ES+) m/z 660 (M+H)+.
