Ethyl 3-[5-(5-{3-cyano-4-[(1-methylethyl)oxy]phenyl}-1,2,4-oxadiazol-3-yl)-1H-indol-1-yl]propanoate (D53) (150 mg, 0.38 mmol) was dissolved in ethanol (25 ml) by warming to 60° C. Allowed solution to cool to RT then added 2N sodium hydroxide (3 ml, 6 mmol). The solution was stirred at RT for 30 minutes. LC/MS showed a single product. Evaporated off the ethanol and filtered off the solid which precipitated out of solution. Mass of title compound as a light tan solid obtained on drying was 50 mg. δH (400 MHz, d6-DMSO) 1.45 (6H, d), 2.67 (2H, t), 4.48 (2H, t), 4.92-4.98 (1H, m), 6.54-6.55 (1H, m), 7.34 (1H, d), 7.38 (1H, d), 7.60 (1H, d), 7.91 (1H, dd), 8.35 (1H, d), 8.41-8.46 (2H, m). MS (ES) C23H20N4O4 requires 416; found 417 (MH+).
