(S,S)-1-Cyclopentyl-3-(2,6-difluoro-3,5-dimethoxy-phenyl)-7-[(5-hydroxymethyl-2-phenyl-[1,3]dioxolan-4-ylmethyl)-amino]-3,4-dihydro-1H-pyrimido[4,5-d]pyrimidin-2-one (0.57 g, 0.95 mmol) was dissolved in 90 mL methanol containing 12 mL 10% HCl acid. The mixture was stirred at room temperature for 9 hours. Adjusted pH=12 by adding 1 M NaOH. Stripped the solvent. The residue was dissolved in methanol. Insoluble NaCl was removed by filtration. The filtrate was evaporated to dryness and the residue was purified by column chromatography (5:1 dichloromethane/methanol) to give 0.275 g (57%) (S,S)-1-cyclopentyl-3-(2,6-difluoro-3,5-dimethoxy-phenyl)-7-(2,3,4-trihydroxy-butylamino)-3,4-dihydro-1H-pyrimido[4,5-d]pyrimidin-2-one as white solid: mp=160° C (dec.); MS (APCI) m/z 510.1, 511.1.
