Reaction Steps
850 mg (1.6 mmol) of N-(2-hydroxyethyl)-4-[({[4-(trifluoromethoxy)phenyl]amino}-carbonyl)amino]-1-[4-(trifluoromethyl)benzyl]-1H-imidazole-2-carboxamide (Example 124) are dissolved in 3.2 ml of absolute DMF under argon, and a few crystals of DMAP and 647 mg (892 μl, 6.4 mmol) of triethylamine are added. While cooling in ice, 755 mg (400 μl, 4.8 mmol) of bromoacetyl chloride are added dropwise. The reaction mixture is stirred at RT for 1.5 h and then poured into cold saturated sodium bicarbonate solution and extracted twice with ethyl acetate. The combined extracts are washed once with water and three times with saturated sodium chloride solution, dried with magnesium sulphate and evaporated in vacuo.
