Reaction Steps

A mixture of 2.11 g of 4-chloro-1-(4-methoxy-phenyl)-butan-1-one (1), 1.60 g of 4-(4-chloro-benzyl)-piperidin-4-ol (5), 1.96 g of K2CO3, and 0.45 g of KI in acetonitrile (50 mL) was stirred and refluxed at 100 to 110° C. for 19 hours under nitrogen atmosphere. After the solvent was removed, ice water was added and the mixture was extracted with ethyl acetate. The organic layer was successively washed with water and brine and dried over MgSO4. After the solvent was removed, the obtained oily residue was purified by silica gel chromatography (chloroform:methanol=20/1 to 10/1) and the purified product was recrystallized from AcOEt/Et2O to afford 1.55 g of compound (6).
