Procedure

A mixture of 2-(4-chloro-2-fluoro-3-methoxyphenyl)-6-(diethoxymethyl)-3-fluoro-N-tritylpyridin-4-amine (2.117 g, 3.44 mmol), CH3CN (15 mL), water (15 mL) and 1 normal (N)H2SO4 (7.5 mL) was placed in an oil bath and heated to 80° C. After 2 h at 80° C., an aliquot of the reaction mixture was partitioned between EtOAc and 10% sodium bicarbonate (NaHCO3) and analyzed by high-performance liquid chromatography (HPLC) and TLC (80/20 hexanes/EtOAc). TLC and HPLC analyses showed only a trace of the starting material remaining (˜2% by HPLC) and essentially one, more polar product.
