Using similar reaction conditions as described in step-ii of example-1, tert-butyl 4-(4-(3-iodo-1-tosyl-1H-pyrrolo[2,3-b]pyridin-5-yl)phenyl)piperazine-1-carboxylate (intermediate 41) (150 mg, 0.227 mmol) was coupled with 3,5-dimethyl-4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)-1-(3-(trifluoromethoxy)benzyl)-1H-pyrazole (Intermediate 44) (135.6 mg, 0.34 mmol) in sodium carbonate (72.4 mg, 0.68 mmol), PdCl2(dppf)Cl2 (8 mg, 0.011 mmol), toluene/ethanol/water (3/5/1 ml) to give 140 mg (76.9% yield) of titled compound. MS: m/z=801.3 (M+1).
