Procedure

4-Chloro-5-methyl-thieno[2,3-d]pyrimidine-6-carboxylic acid methyl ester (1.25 g), tert-butyl 3-(2-amino-5-fluorophenoxy)butylcarbamate (1.7 g) and p-toloulsulfonic acid (175 mg) was dissolved in dioxane (30 ml) and the solution was heated under reflux overnight. Then the reaction mixture was concentrated in vacuo. The residue was treated with trifluoracetic acid 25% in dichloromethane and stirred at room temperature for 1 h. The solution was concentrated in vacuo and the residue was dissolved in dichloromethane and washed with sodium bicarbonate. The organic phase was separated, dried and concentrated in vacuo. The residue was triturated with Et2O and the precipitate was filtered.
