Reaction Steps
To a solution of tert-butyl 4-chloro-2-propyl-2H-pyrazolo[3,4-c]quinolin-1-ylcarbamate (722 mg, 2.00 mmol) in THF (5 mL) was added ethyl iodide (624 mg, 4.0 mmol). Sodium hydride (120 mg of a 60 wt % dispersion in mineral oil, 3.00 mmol) was added, and after 40 min, more ethyl iodide (624 mg, 2.00 mmol) was added, and the reaction was stirred overnight. Saturated ammonium chloride was added, and the aqueous layer was extracted with methyl tert-butyl ether (3×). The combined organic layers were dried (magnesium sulfate), filtered, concentrated, and purified by prep HPLC (silica cartridge, eluting with ethyl acetate in hexane) to afford 718 mg of tert-butyl 4-chloro-2-propyl-2H-pyrazolo[3,4-c]quinolin-1-yl(ethyl)carbamate as a white solid.
