Reaction Steps
To a stirred solution of methyl (4-nitro-1-{[(7S)-2′-oxo-1′,2′,6,8-tetrahydrospiro[cyclopenta[g]quinoline-7,3′-pyrrolo[2,3-b]pyridin]-2-yl]methyl}-1H-indol-3-yl)acetate from Step A (22 mg, 0.041 mmol) in H2O (0.5 mL) and EtOH (0.5 mL) was added sodium hydrosulfite (ca. 85%; 108 mg, 0.53 mmol). The reaction mixture was stirred at ambient temperature for 1 h, then at 50° C. for 90 min. The reaction mixture was allowed to cool, more sodium hydrosulfite (ca. 85%; 55 mg, 0.27 mmol) was added, and the resulting mixture was heated to 70° C. for 4 h. 3 N hydrochloric acid (0.55 mL, 1.65 mmol) was added and the solution was heated to 70° C. for 18 h. EtOH (1 mL) was added and heating was continued at 100° C. for 24 h, then the cooled mixture was concentrated in vacuo. The residue was purified by HPLC using a reversed phase C18 column and eluting with a gradient of H2O:CH3CN:CF3CO2H—90:10:0.1 to 5:95:0.1. The pure, product-containing fractions were combined and concentrated to give the title compound. MS: m/z=472 (M+1). HRMS: m/z=472.1764; calculated m/z=472.1768 for C29H22N5O2.
